The club will be closed for more than a month so it can undergo a refurbishment before it reopens in early March as a Popworld.
Images from inside the project show work crews have made a swift start to the works, stripping most of the floor and panelling ahead of the major rebrand.
Popworld is a growing chain of clubs, with sites up and down the country including one in Chelmsford.
It describes itself as a late night bar “with a difference” boasting bright colours, sweet cocktails, light-up dancefloors and back-to-back "cheesy" pop tunes from the 90s and noughties.
Jade Bailey, 29, of Southend, said: "I love S Club 7, Steps, Britney Spears and all those kind of bands so I cannot wait for it to reopen as Popworld.
"From the pictures it looks like they are really changing everything about the interior so I am very excited to see the finished product."
Michelle Chapman, 32, said: "It's a shame to say goodbye to Rhythmn Room as I have some great memories of that place, including my aunt's hen doo.
"But I am a massive fan of pop music so I can't lie and say I'm not excited."
It is not the first time the venue has rebraned. Rhythm Room itself replaced the old Dick De Vignes nightclub, a longstanding venue in the city centre that ran for roughly 17 years.
Rhythm Room posted on Facebook: “Refurbishment works are well underway as we get ready to bring you a brand new nightclub experience to Southend city.
“This is gonna Pop off!”
A spokesman for Rhythm Room added: “We are excited to announce that the Popworld party is coming to Southend.
“Rhythm Room will be closing for a few weeks, while it undergoes a transformation.
“The venue will be reopening as the iconic Popworld in early March, keep your eyes peeled for updates and be sure to follow our news on social media.”
